    Abstract / Zusammenfassung

In the present work an orthotopic pancreaticoduodenal transplant model was developed and the metabolic consequences of this technique were examined.Twelve inbred rats had undergone pancreaticoduodenectomy before receiving an orthotopic duodenopancreas transplant with venous outflow via portal vein and enteric drainage of the exocrine secretion. Twelve laparatomized rats served as control. Three months after the operations all rats received indwelling catheters in the carotid artery, the jugular vein, and in the mouth. Each rat underwent an oral and and Jsoglycemic" intravenous glucose tolerance test with arterial blood sampling for plasma glucose, insulin, c-peptide, and GIP determinations. The results show no change in the endocrine function after transplantation. Slightly lower incremental plasma glucose after oral glucose in the transplanted animals likely esults from decreased intestinal glucose uptake due to denervation. Unchanged fecal fat and chymotrypsin show absence of intestinal malabsorbtion of lipids and suggest preserved exocrine pancreatic function as well.Orthotopic transplantation therefore qualifies as treatment modality for disease states of the pancreas wich are surgically untreatable without pancreatectomy.
